The San Francisco 49ers enter Week 16 at 10–4, firmly in playoff position with three games remaining. Read on for 49ers key players to watch as San Francisco faces the struggling Indianapolis Colts. With playoff positioning and momentum on the line, the 49ers will need contributions from both offense and defense to continue their winning streak.
A 49ers victory on Monday Night Football would all but extinguish the Colts playoff hopes. The two teams are trending in opposite directions. The 49ers are seeking their fifth straight win, while the Colts (8–6) are trying to snap a four-game losing streak. In Week 14, Colts quarterback Daniel Jones suffered a torn Achilles that will sideline him for the remainder of the season, dealing a significant blow to Indianapolis’ late-season momentum.

Game implications
Jones’ absence and the Colts’ second-half slump have dulled some of the buzz surrounding this matchup. Yet, the game remains meaningful for both sides. The 49ers are still battling for the NFC West title and improved playoff seeding, while the Colts cling to hopes of a late-season push to sneak into the postseason. The heightened stakes, Monday Night Football spotlight, star power on both rosters and a road environment suggest this will be a tougher test for San Francisco than last week’s win over the Tennessee Titans.
If the 49ers want to extend their winning streak in their final matchup against an AFC opponent in 2025, the following players will need to play key roles.
49ers Key Players to Watch: Offense
Against Tennessee, the 49ers offense found balance, producing through both the air and on the ground. That balance will remain crucial moving forward, as coach Kyle Shanahan’s offense looks to keep defenses guessing rather than placing the burden solely on quarterback Brock Purdy or running back Christian McCaffrey.
Second-year wide receiver Ricky Pearsall turned in arguably his best performance of the season last week, catching six passes for 96 yards. However, he sustained knee and ankle injuries. Even if Pearsall plays, he may not be at full strength.
That could open the door for increased opportunities for Jennings and Kittle, both of whom found the end zone against the Titans. Against an average Colts defense, Purdy should look to Kittle early and often for chunk gains. Kittle will also have extra motivation to assert himself as one of the league’s premier tight ends with the Colts talented rookie tight end Tyler Warren lining up on the opposite sideline.
49ers Key Players to Watch: Defensive Front
San Francisco’s pass rush has been a season-long concern. The 49ers have just 16 sacks through 14 games, fewer than Cleveland Browns star Myles Garrett, who has 21.5 on his own.
This matchup presents an opportunity for the 49ers’ defensive front to make an impact. With Jones sidelined, the Colts turned to veteran Philip Rivers, who made his NFL return last Sunday. The 44-year-old San Diego/Los Angeles Chargers legend will start for the second straight game on Monday night. Rivers has never been a mobile quarterback, which could allow San Francisco’s defensive line to collapse the pocket and generate pressure.
If defensive linemen Bryce Huff, Clelin Ferrell, and Sam Okuayinonu consistently win their matchups, it would help get the Colts’ offense off the field and limit Jonathan Taylor’s impact.
49ers Linebackers
The timing could not be better for the 49ers to get linebacker Tatum Bethune back on the field. After returning to limited practice this week, Bethune is trending toward a return following a three-game absence.
His presence will be critical against a Colts offense likely to lean heavily on Taylor. Bethune and fellow linebacker Dee Winters must be strong in run support to prevent Taylor from becoming the latest running back to break loose against San Francisco’s defense.
A strong showing from Bethune and the defensive front could be the key to keeping the 49ers rolling and sending a statement on the road in Indianapolis.
